No, they can be used again.
The Dainese ($$$
needs to be sent back to the distributor to be rearmed. It's a very nice system and relies on a CPU and inertia sensors for deployment. This is what's used on the racing circuit.
The Helite is much simpler. It's connected to your bike via quick-release tether and uses a single-use replaceable CO2 cartridge which is easily swapped out after deployment. The cartridges for my vest cost $25.
Other inexpensive brands are beginning to appear on the market. I'm not sure how they work, but they're probably similar to the Helite.
